Materials and Methods

Cell Culture and Transfection

Human HepG2 cells were obtained from the ATCC (Manassa, VA, Brazil). Cells were cultured in Dulbecco’s modified Eagle’s medium (DMEM, Gibco, Brazil) supplemented with 10% fetal bovine serum (FBS, Gibco) and 1% penicillin/streptomycin (Gibco). For cell culture, cells were seeded in a 12-well plate at a density of 5,000 cells/well. After 24-h incubation at 37°C, cells were plated in 1 ml DMEM with the exception of FBS, and the medium was changed every 4–6 h. Cell confluence was achieved with Dulbecco’s Modified Eagle’s Medium with FBS (Gibco), and cells were allowed to recover in DMEM for 3 days.

The blots were then visualized using EITHER optics (Nanjing Quaizhou, China) and the bands were then quantified using Quantity one version (Bio-Rad Laboratories, Hercules, CA, USA). The intensity of the bands was normalized to the intensity of the corresponding β-actin antibody.
