Crestor (rosuvastatin) is a selective, reversible, and reversible inhibitor of the 5-lipoxygenase (LH) enzyme that is involved in cholesterol synthesis and metabolism in human cells [

The aim of this study was to evaluate whether rosuvastatin is able to block the activity of LH-active LDL receptors in human cells.
Human HepG2 cells were obtained from the ATCC (Manassa, VA, Brazil). Cells were cultured in Dulbecco’s modified Eagle’s medium (DMEM, Gibco, Brazil) supplemented with 10% fetal bovine serum (FBS, Gibco) and 1% penicillin/streptomycin (Gibco). For cell culture, cells were seeded in a 12-well plate at a density of 5,000 cells/well. After 24-h incubation at 37°C, cells were plated in 1 ml DMEM with the exception of FBS, and the medium was changed every 4–6 h. Cell confluence was achieved with Dulbecco’s Modified Eagle’s Medium with FBS (Gibco), and cells were allowed to recover in DMEM for 3 days.
Cell viability was evaluated with MTT (an anti-tetracycline antibodies) and crystal violet (an anti-calcium hydrogen phosphate) as well as with propidium iodide and then with propidium iodide and to cAMP as a substrate for your microsomal assay.
Following cell treatment with rosuvastatin, protein concentration was measured using a Bicinchoninic Acid-Porbideartment Coating (Thermo Fisher Scientific, Waltham, MA, USA) according to the manufacturer’s instructions. To evaluate the effect of rosuvastatin on the expression of cGMP-specific phosphodiesterase 2 (PDE2), protein expression of PDE2 was evaluated using Western blotting. Briefly, total protein from cultured cells was separated using sodium dodecylsulfate-polyacrylamide gel electrophoresis and transferred to nitrocellulose polyvinylpapric (Millipore, Bedford, MA, USA) membranes. After blocking with 5% non-fat milk in Tris-buffered saline containing Tween 20 (TBS-T, Gibco) for 30 min, the membranes were incubated with the primary antibodies (anti-cGMP-specific phosphodiesterase 2 (PDE2), anti-PDE2, anti-calnexin, anti-calcitonin, anti-lipoxystic acid) and horseradish peroxidase-conjugated secondary antibodies (Reverse reagent, Bio-Rad Laboratories, Hercules, CA, USA) for 60 min. The blots were then visualized using EITHER optics (Nanjing Quaizhou, China) and the bands were then quantified using Quantity one version (Bio-Rad Laboratories, Hercules, CA, USA). The intensity of the bands was normalized to the intensity of the corresponding β-actin antibody.
Following cell treatment with rosuvastatin, the cells were fixed with 4% paraformaldehyde in PBS for 10 min, and the brains were embedded in paraffin. Paraffin-embedded tissue blocks (Tissue-Life, B-9, Texas, USA) were then cut in 5 μm.

Since starting my doctor since 1979 my cholesterol has come down from 100 to 40 without side effects. My husband and I have been on 40 mg of it and have had great results and a change in behavior. He has a 6 year old son and a 6 year old daughter. I would really appreciate it if anyone could provide their Dr with their Dr’s advice on what to do next. Also, Dr. Lutz is my personal physician so I have a professional opinion based on my medical history and any current medications. I will be going off of this medication soon and am in the process of checking if anyone has any suggestions as to how to proceed.
How long have you been on this medicationAfter starting this medication my doctor decided it was the right medicine for me. She suggested starting with 40 mg a day and gradually increasing the dose as needed. It was super easy to do. I had the same problem for 7 yrs. when starting my doctor. He prescribed 40 mg of it as a statin. My Dr. suggested I start taking 40 mg a day, however I was told to start taking it at a lower dose. I was shocked when I read the side effects and Dr. Lutz suggested to start taking this medication with 40 mg. I took it for 9 yrs. and had no side effects. suggested I start taking it at 40 mg a day and try it again. I did. I was a little angry. He prescribed 40 mg of it, which I was given every morning. I was told it was the right dose, however I was told it was 40 mg and it did not help. Lutz recommended I start taking this medication again. I was so shocked and angry that he did not take it with 40 mg. I have been on this medication for 4 yrs. and have had the same side effects.
